Modular biotechnological equipment of the "OKA-01" series

Function

Modular biotechnological equipment of the "Oka-01" series is designed for cultivation of bacterial, yeast and mycelial forms of microorganisms.

It consists of several autonomous modules

  • Sterilization module;
  • Fermentation molule;
  • Interactive control module.

This equipment can be used for both fundamental and applied studies in biotechnology and also for experimental-industrial and industrial manufacture of products.

Sterilization module contains

Continuously working steam generator PG-20, which produces saturated water steam of given temperature and pressure. The steam generator provides saturated water steam without gas impurities and stabilizes the level of the vaporised water. It prevents blowout of droplet water with steam flow from the steam generator and allows refilling of water to the steam generator by means of a thermal pump.

Sterilizator of solutions SR-10, which performs continuous dosed sterilization of liquid nutrient medium at given temperature, pressure and the number of sterilization cycles. The sterilizer performs accelerated sterilization due to intensive and controlled agitation of the sterilized nutrient medium by means of saturated water steam, provides forced condensation of the steam with the formation of distilled water and sterilizes concentrated nutrient medium followed by a dilution by distilled water.

Sterilizer of gases SG-3, which has three channels of membrane filtration of gases and sterilizes aeration air, exhaust and technological gases.

Autoclave AK-25 is designed for sterilization of glass ware, tools and titrating solutions. The use of the autoclave reduces the time of inactivation of microorganisms during sterilization due to the cyclic steam flow in the autoclave.

Fermentation molules

Fermentation molules are produced with mechanical or pneumohydraulic agitation.

Fermentation modules with mechanical agitation possess a stainless steel bioreactor of a given volume (MF-10T and MF-100T for 10 L and 100 L bioreactors) and includes

  • Turbine agitator with magnetic drive;
  • Steam-sterilized connector for sampling and connection with external units in conditions of aseptic cleanness;
  • Pressure seals for accommodation of sensors of temperature, pH, dissolved oxygen, and delivery of titrating solutions;
  • Heat exchanger;
  • Steam condenser;
  • A dispencer for delivery of nutrient media to the bioreactor and another one for discharge of culture liquid;
  • A cabinet for the arrangement of the equipment.

The design of the agitation element and the aeration system practically totally prevents foam formation during the aeration and agitation of culture liquid. It results in an increase in load factor of fermentors up to 0.8. It allows as well keeping stable the level of the culture liquid in the bioreactor and guarantees its antiseptic conditions at any modes of fermentation.

Fermentation molules with pneumohydraulic agitation

Fermentation molules with pneumohydraulic agitation are designed for the work in cascade mode and has stainless steel bioreactors of given volume (0.5 to 3 L). The module includes

  • Three bioreactors;
  • Steam-sterilized connectors for sampling and connection with external units in conditions of aseptic cleanness;
  • Pressure seals for accommodation of sensors of temperature, pH, dissolved oxygen, and delivery of titrating solutions;
  • Heat exchanger;
  • Steam condenser;
  • Bacterial air and discharge gas filters;
  • Transfer chamber for aseptic discharge of culture liquid

The interactive control module

The interactive control module provide full information about the sate of running processes by means of symbolic circuits, graphic and symbol display and is used to control multiphase processes.

The module contains

  • Micropocessor-based controller;
  • A computer;
  • A set of computer programs for: sterilizator of liquid nutrient medium, autoclave, fermentation module. This program controls the following operations: bioreactor washing, bioreactor sterilization, realization of flow fermentation processes.

Feasible configurations of systems consisting from the abovementioned modules are shown in Figures 1 to 3.
