Conformational changes in GPCR signaling

19 ноября 2015 г в Лаборатории новых методов в биологии выступил с докладом на тему «Conformational changes in GPCR signaling» российский ученый-исследователь, бывший сотрудник ИБП РАН Дмитрий Вепринцев.

В настоящее время Дмитрий Борисович является руководителем проекта Лаборатории биомолекулярных исследований (Laboratory of Biomolecular Research) института Пола Шеррера (Paul Scherrer Institut) в Швейцарии и занимается изучением рецепторов, действующих через активацию G-белка (от англ. G-protein coupled receptors, GPCR). GPCRs выполняют функцию активаторов внутриклеточных путей передачи сигнала, приводящими в итоге к клеточному ответу.

В своем докладе Вепринцев подчеркнул важность изучения GPCRs: «G protein-coupled receptors (GPCRs) are membrane proteins responsible for transmitting the signal carried by a variety of extracellular stimuli, such as hormones or signalling peptides, across the cellular membrane. Upon binding of ligands they interact with intracellular G proteins, receptor kinases and arrestins activating several signalling pathways. Due to the importance of GPCRs in cell physiology they are targeted by numerous pharmacological ligands. Some of such ligands preferentially activate one of the pathways and are of particular interest for therapeutic application. One of the most important questions is to understand how ligands can induce distinct conformations in GPCRs and activate different downstream signalling pathways. The research of my group is focused on identification of the number of distinct conformational states in GPCRs and G proteins and dynamics of their exchange, structural characterisation of ligand-induced conformational changes and their effects on signalling. To achieve this goal we use a combination of protein engineering, scanning mutagenesis, biophysics, x-ray crystallography and NMR».
